N3-kethoxal
N3-kethoxal is a small, membrane-permeable probe, which selectively binds to un-paired guanine residues within a cellular environment and then provides a bio-orthogonal handle for click chemistry reactions. Depending on how the azide group is utilized, N3-kethoxal is amenable to various nucleic acid probing applications. The N3-kethoxal platform can be utilized to probe RNA secondary and tertiary structure, map single stranded DNA regions in the genome, explore RNA-RNA interactions, and identify protein-RNA interactions. Compared with current nucleic acid probes, N3-kethoxal exhibits low toxicity, as well as high specificity, resolution, and reproducibility, providing a novel click chemistry platform for in vitro and in vivo analyses of nucleic acid structure and proximity interactions.
