Background

glycochenodeoxycholate (CAS No.: 640-79-9) is a glycine-conjugated bile acid derived from chenodeoxycholic acid in the liver and represents a physiologically relevant bile salt involved in lipid emulsification and intestinal fat absorption, functioning as a detergent as well as a cholagogue and choleretic agent. In biomedical research, it is widely utilized as a model bile acid to investigate hepatobiliary physiology and bile acid–mediated cytotoxicity, particularly in the context of cholestatic liver injury. Mechanistically, glycochenodeoxycholate has been shown to disrupt cellular homeostasis in hepatocytes by inhibiting autophagosome formation, impairing lysosomal proteolysis, and increasing lysosomal pH, ultimately leading to apoptosis; it also modulates signaling pathways such as STAT3, contributing to enhanced stemness and chemoresistance in hepatocellular carcinoma models. In vitro studies indicate that its biological effects, including cytotoxicity and modulation of autophagy markers, typically occur in the low to mid micromolar range, depending on cell type and experimental conditions.
