IdeZ Protease (IgG-specific)
IdeZ protease (IgG-specific), also known as immunoglobulin-degrading enzyme from Streptococcus Equi subspecies zooepidemicus, also known as immunoglobulin-specific IdeZ protease, is an immunoglobulin IgG degrading enzyme (Immunoglubulin G-degrading enzyme). The protease has extremely high substrate specificity and recognizes only IgG, which is digested at a specific site in the hinge region under the antibody, hydrolyzing IgG into intact F(ab')2 and Fc fragments. IdeZ protease (IgG-specific) is mainly used as a tool enzyme for the structural characterization of antibody drugs or antibody fusion protein drugs, to prepare antibodies containing only F(ab')2 fragments or Fc fragments of antibodies, and can also be used to remove heavy chains from the interference of target protein detection during Western detection after immunoprecipitation.
IdeZ protease (IgG-specific) effectively cleaves human IgG1, IgG2, IgG3, and IgG4, as well as rabbit, rat, monkey, sheep, humanized and chimeric IgG, and Fc fusion proteins and ADCs, but not mouse IgG1/IgG2b, porcine, bovine, or goat IgG, and non-IgG isotype immunoglobulins, including IgA, IgM, IgD, and IgE. At the same time, extended incubation time can also cleave mouse IgG2a and IgG3. Compared with IdeS Protease (IgG-specific), the digestion activity of IdeZ protease (IgG-specific) on rats, mouse IgG2a and IgG3 was significantly improved.
This product is a recombinant protease obtained from the expression of Escherichia coli, which has the
characteristics of high purity and strong enzymatic digestion activity. The protease contains the His tag, which can be subsequently removed by nickel columns. Using this product, 100 reactions can be performed in 1 kU using 20 μL as an example.
