Verubecestat (MK-8931)
Verubecestat (MK-8931) (CAS 1286770-55-5) is an orally bioavailable small-molecule inhibitor, functioning as a potent beta-site amyloid precursor protein-cleaving enzyme 1 (BACE1) inhibitor in neuronal tissues and exhibiting strong antagonistic activity toward BACE1-mediated amyloid-beta (Aβ) production. Additionally, it acts to reduce the formation of neurotoxic Aβ peptides by blocking the initial step in the amyloidogenic pathway.
In preclinical experimental contexts, Verubecestat (MK-8931) significantly suppresses Aβ peptide generation with an IC50 value of 2.2 nM, tested against in vitro human neuronal cell lines expressing amyloid precursor protein (APP). It can also decrease Aβ40 production in a dose-dependent manner, as demonstrated in cellular models relevant to Alzheimer's disease pathology. Furthermore, Verubecestat (MK-8931) has been shown to attenuate Aβ accumulation in transgenic mouse models of Alzheimer's disease, highlighting its efficacy in mitigating amyloid plaque development through BACE1 inhibition.
In clinical application contexts, Verubecestat (MK-8931) is widely used as a pharmacological tool for the investigation of BACE1-targeted therapeutic strategies in Alzheimer’s disease. Its potent inhibitory effects on Aβ formation render it an important compound in both basic research and pharmaceutical development focusing on neurodegenerative disorders characterized by amyloid pathology.
