Sulfo-NHS-LC-D-Desthiobiotin
Sulfo-NHS-LC-D-Desthiobiotin is a sulfonated, long-chain amine-reactive biotin labeling reagent that is activated into Sulfo-NHS with a long-chain (LC) spacer arm esters to covalently label primary amines (-NH2) of proteins or other molecules by dethiobiotin groups. The desthiobiotin tag binds to streptavidin and other biotin-binding proteins with high specificity but is easy to elute under mild conditions. This reagent is an effective alternative to Sulfo-NHS-LC-Biotin (A8003) for avidin-biotin techniques where native elution of tagged proteins is required. The negatively charged Sulfo-NHS group prevents this reagent from penetrating the cell membrane, and therefore, Sulfo-NHS-LC-D-Desthiobiotin can be used to limit the cell surface proteins that label intact cells.
Features:
- Desthiobiotin: Readily elutes from streptavidin, which is ideal for affinity purification applications.
- Amine reactivity: Reacts with primary amines (-NH2) such as lysine side chains or the amino terminus of polypeptides.
- Solubility: Sulfo-NHS groups improve water solubility.
- Cell surface labeling: Negatively charged reagents do not penetrate the cell membrane and only modify the surface proteins of the whole cell.
- Protein Labeling: Label antibodies or proteins in purified or pooled samples to achieve high recovery in sedimentation assays with streptavidin beads.
