Pemetrexed

Pemetrexed (CAS No.: 137281-23-3) is a synthetic antifolate and guanine-derived small molecule widely utilized in biochemical and pharmacological research as a multi-target inhibitor of folate-dependent enzymes involved in nucleotide biosynthesis. It exerts its activity primarily through potent inhibition of thymidylate synthase, dihydrofolate reductase, and glycinamide ribonucleotide formyltransferase, key enzymes in the de novo synthesis of thymidine and purine nucleotides, thereby disrupting DNA and RNA synthesis pathways. The polyglutamated intracellular forms of pemetrexed exhibit high affinity for these targets, with inhibitory constants reported in the nanomolar range, reflecting strong enzyme binding and sustained intracellular retention.
