N-Acetyl-L-leucine
N-Acetyl-L-leucine (CAS No.: 1188-21-2) is an orally bioavailable, brain-penetrant endogenous metabolite and an acetylated derivative of the essential amino acid L-leucine, widely studied for its neuroprotective and metabolic regulatory properties in biomedical research. As the active L-enantiomer of N-acetyl-leucine, it has been shown in preclinical models to attenuate neuronal cell death and neuroinflammation, particularly within cortical tissues, and to improve lysosomal function and cellular metabolic homeostasis, suggesting a role in modulating pathways associated with cellular stress responses and organelle dysfunction. Although its precise molecular targets remain incompletely defined, its effects are thought to involve normalization of disrupted metabolic signaling and enhancement of neuronal resilience under pathological conditions. In vivo studies further demonstrate its ability to improve functional recovery and postural compensation in models of vestibular injury, supporting its relevance in neurophysiological regulation. N-Acetyl-L-leucine is therefore broadly applied in studies of neurodegenerative disorders, lysosomal storage diseases such as Niemann-Pick type C, and traumatic brain injury, as well as in screening platforms aimed at identifying modulators of neuroinflammation and metabolic dysfunction. Experimental usage in cell-based and animal models typically employs concentration or dosing ranges tailored to specific assay systems, often spanning from low micromolar to higher exposure levels depending on the biological context and study design.
