Lipid nanoparticles (LNPs) represent a leading class of non-viral nucleic acid delivery vectors, widely recognized for their efficient encapsulation and targeted delivery capabilities. This classic SM-102 type mRNA-LNP preparation kit is engineered to enable reliable small-scale formulation of mRNA-loaded LNPs.
LNPs are formed via rapid micro-mixing of a lipid-containing ethanol phase with an mRNA-containing acidic aqueous phase. Under the acidic conditions provided by the kit's citrate buffer, the ionizable lipid SM-102 undergoes protonation, acquiring a positive charge that electrostatically binds to negatively charged mRNA strands, driving efficient nucleic acid encapsulation within the nanoparticle core. Following system neutralization, the LNP surface transitions to a near-neutral state, minimizing non-specific interactions while enhancing biocompatibility.
By encapsulating mRNA within LNPs, the system effectively shields the labile nucleic acid from degradation by extracellular nucleases, promotes cellular internalization via endocytic pathways, and facilitates cytoplasmic release to enable efficient translation of the encoded protein.
