7-Deaza-dGTP is a nucleotide analog where the nitrogen atom at position 7 of the deoxyguanosine ring is replaced by a carbon atom. This modification eliminates the major site for chemical cleavage and prevents the formation of G-quartet structures in GC-rich regions.
It is specifically designed to overcome compressions and stops in DNA sequencing by reducing secondary structures. This results in clearer, more interpretable sequencing ladders, especially for templates with high GC content.
