Background

ddTTP (2',3'-dideoxythymidine triphosphate) is a thymidine nucleotide analog utilized in molecular biology research. Structurally lacking hydroxyl groups at both the 2' and 3' ribose positions, it acts as a chain terminator during DNA synthesis catalyzed by DNA polymerases. Incorporation of ddTTP into a growing DNA strand prevents subsequent nucleotide addition, effectively terminating elongation. Due to this property, ddTTP is commonly employed in Sanger sequencing and primer extension assays to accurately determine DNA sequences and identify mutations.
