4-Methoxychalcone-1
4-Methoxychalcone (4-MC, CAS No. 959-33-1) is a plant-derived chalcone that can also be synthesized via Claisen-Schmidt condensation. 4-MC exhibits dual functions: it promotes adipocyte differentiation and selectively eliminates senescent endothelial cells. At 5 μM, 4-MC significantly enhances 3T3-L1 preadipocyte differentiation by activating the PPARγ pathway and upregulating adiponectin, aP2, and FAS, while counteracting TNF-α-induced inhibition and reducing inflammatory markers (IL-6, PAI-1, MCP-1). For anti-aging, 4-MC shows potent activity against senescent HAECs (IC₅₀ = 3.42 μM, SI = 3.4), outperforming fisetin, and exhibits low cytotoxicity to IMR90 cells (IC₅₀ = 24.32 μM, SI = 2.1), indicating endothelial specificity. These properties highlight 4-MC’s potential in metabolic syndrome and cardiovascular aging research.
