GSK J4 free base
GSK J4 free base (CAS 1373423-53-0) is a small-molecule inhibitor targeting the histone demethylases JMJD3/KDM6B and UTX/KDM6A. It functions by preventing the demethylation of histone H3 lysine 27 (H3K27me3/me2), resulting in epigenetic modulation. In vitro assays demonstrate that GSK J4 inhibits JMJD3/KDM6B and UTX/KDM6A with micromolar potency. In cellular studies, it suppresses inflammatory responses, notably reducing TNF-α secretion in LPS-stimulated human primary macrophages, and induces apoptosis associated with endoplasmic reticulum stress. GSK J4 is commonly employed in epigenetic and inflammation-related disease research.
