Glutathione Peroxidase Activity Colorimetric Assay Kit
Glutathione peroxidase (GPx) is an important antioxidant enzyme that catalyzes the reduction of hydrogen peroxide and organic peroxides to water or alcohols using reduced glutathione (GSH), thereby eliminating peroxides and protecting cells from oxidative damage. GPx is widely distributed in various tissues, and its activity level is closely associated with pathophysiological states, making the detection of GPx activity highly significant for disease research.
This kit indirectly determines GPx activity by monitoring NADPH consumption. In the coupled reaction, GPx catalyzes the oxidation of GSH to GSSG while reducing peroxides; glutathione reductase (GR) then regenerates GSH from GSSG using NADPH. The decrease in NADPH absorbance at 340 nm reflects GPx activity. The kit uses t-Bu-BOOH as a substrate, which is not metabolized by catalase, effectively avoiding interference from catalase activity. Additionally, t-BOOH is selectively reduced by selenium-dependent GPx (the predominant form in cells), ensuring specific detection of this isoform. This kit is easy to use and suitable for various sample types, including tissues and cells.
