Denifanstat (TVB-2640) is an orally effective selective fatty acid synthetase (FASN) inhibitor. Fatty acid synthetase plays a key role in lipid metabolism of tumor cells. Inhibition of this enzyme can effectively block lipid synthesis of tumor cells and inhibit tumor growth. Denifanstat has high selectivity and activity, with IC50 of 0.052 μM for FASN and EC50 of 0.072 μM at cellular level, indicating that Denifanstat can effectively inhibit the target enzyme activity and play an anti-tumor role at a low concentration. At present, the drug has entered the clinical research stage, showing good safety and initial efficacy, and is expected to become a new targeted drug for the treatment of a variety of cancers.
