Concanavalin A
Concanavalin A (CAS No.: 11028-71-0) is a plant-derived lectin isolated from Canavalia ensiformis (jack bean) that selectively and competitively binds α-D-mannose- and α-D-glucose-containing glycoconjugates, making it a widely used probe in glycobiology and immunology. Functionally, it acts as a potent T cell mitogen that stimulates lymphocyte activation, proliferation, and cytokine production through crosslinking of cell-surface glycoproteins and modulation of downstream signaling pathways. In addition to its immunostimulatory properties, Concanavalin A induces programmed cell death via mitochondria-dependent apoptosis and autophagy, characterized by loss of mitochondrial membrane potential, apoptotic nuclear morphology, and perturbation of intracellular calcium homeostasis, including depletion and oscillatory changes in apoptotic cells. In vitro, its biological effects are typically observed at micromolar to tens of micrograms per milliliter concentrations, depending on cell type and assay conditions. In vivo studies demonstrate immunomodulatory and antitumor activities, including inhibition of tumor growth in murine models at appropriate dosing regimens. Concanavalin A can also be immobilized on matrices such as agarose for affinity purification of glycoproteins and is employed in biochemical assays, immune cell activation studies, and glycan profiling, as well as in biosensing applications such as glucose monitoring. Due to its cytotoxic, hepatotoxic, and teratogenic potential at higher exposures, experimental concentrations or doses should be carefully optimized according to specific research objectives.
