C34 (CAS 40592-88-9) is a small molecule inhibitor that modulates Toll-like receptor 4 (TLR4) signaling. Structurally classified as a 2-acetamidopyranoside derivative, C34 selectively suppresses TLR4-mediated inflammatory signaling in macrophages and enterocytes, without affecting TLR2 or TLR9 pathways. In vitro studies indicate significant inhibition at concentrations around 10 μM. In vivo, administration of C34 at doses of approximately 1 mg/kg reduces systemic inflammatory responses in animal models of endotoxemia and necrotizing enterocolitis. Additionally, C34 down-regulates basal and LPS-induced TNFα expression and iNOS levels in human intestinal tissues from necrotizing enterocolitis patients, emphasizing potential therapeutic relevance in inflammatory research.
