Background

Mavorixafor hydrochloride (CAS No. 880549-30-4) is the hydrochloride salt form of Mavorixafor (CAS No. 558447-26-0, Catalog No.: A3173). As an oral selective C-X-C chemokine receptor 4 (CXCR4) antagonist, its core target is CXCR4 (blocking the downstream signals of its overactivation). Its biological activity is mainly reflected in inhibiting the CXCR4/CXCL12 signaling axis, improving bone marrow cell migration disorders in patients with WHIM syndrome, significantly increasing neutrophil and lymphocyte counts (the time to reach the target neutrophil count is 15.0 hours vs. 2.8 hours with placebo; the time to reach the target lymphocyte count is 15.8 hours vs. 4.6 hours with placebo), and reducing the annual infection rate by 60%. Meanwhile, it can serve as a potential therapeutic drug for Waldenström's Macroglobulinemia (WM) associated with CXCR4 mutations, and combined use with ibrutinib (CAS No. 936563-96-1, Catalog No.: A3001) is expected to enhance therapeutic efficacy. It has good safety profiles: common adverse reactions are mild to moderate gastrointestinal symptoms and skin disorders, with no serious treatment-related adverse events or cases of treatment discontinuation.
