3-Methylindole (CAS No.: 83-34-1) is an endogenous tryptophan-derived metabolite, also known as skatole, that is produced by intestinal microbiota and naturally occurs in the gastrointestinal tract of mammals as well as in certain plants, classifying it as a microbiome-associated heterocyclic aromatic compound of relevance to gut physiology and toxicology. In biomedical research, it is widely used as a probe molecule to investigate host–microbiome interactions and intestinal epithelial responses, with established activity involving activation of the aryl hydrocarbon receptor (AhR) and downstream signaling pathways such as p38 MAPK, leading to modulation of gene transcription and induction of apoptosis in intestinal epithelial cell models including Caco-2 cells.
