1,2-Dioleoyl-sn-glycero-3-PE (DOPE) is a core helper phospholipid for cationic liposomes. After cellular internalization, it triggers membrane fusion in the acidic endosomal microenvironment, facilitating the release of encapsulated nucleic acids from endosomes into the cytoplasm and thereby boosting transfection efficiency. It is commonly formulated with cationic lipids and DSPE-PEG, and widely applied in the development of LNP-based nucleic acid delivery systems, in vitro transfection reagents, genetic vaccine carriers, as well as anti-tumor nanomedicines.
