Semaglutide
Semaglutide is a long-acting GLP‑1 receptor agonist. Its structure is modified with a fatty acid and binds to albumin, with a half-life of about 1 week. It activates GLP‑1R on pancreatic β cells to promote glucose-dependent insulin secretion, while inhibiting glucagon from α cells, slowing gastric emptying, and acting on central appetite centers such as the hypothalamus to reduce food intake and body weight. Related pathways include cAMP/PKA, PI3K/AKT, and others. It is used for the treatment of type 2 diabetes and obesity, and cardiovascular outcomes trials have shown a reduction in MACE risk.
