Background

OSU-T315 (CAS No.: 2070015-22-2) is a synthetic small molecule classified as an integrin-linked kinase (ILK) inhibitor and signaling pathway research probe. Originating from an academic drug discovery program, it is mainly used to dissect kinase-regulated intracellular signaling and cell fate regulation in the contexts of tumor biology and immunobiology. It weakens PI3K–AKT signal transduction by inhibiting ILK and promoting dephosphorylation of AKT at Ser473, while also modulating previously established AKT substrates (such as GSK-3β and myosin light chain); in addition, it can block AKT translocation to cholesterol-rich lipid rafts, inhibit its full activation, and, in certain cellular contexts, trigger caspase-dependent apoptosis through an ILK-independent pathway, often accompanied by induction of autophagy. Together, these processes contribute to its antiproliferative activity in multiple tumor-derived cellular systems. Reported in vitro potency is generally in the submicromolar to low micromolar range, consistent with effective pathway engagement under standard experimental conditions. Typical applications include dissecting AKT-centered survival and stress signaling in cultured solid tumor and hematologic cell models, monitoring phosphorylation changes in biomarkers such as AKT Ser473, ERK, and GSK-3β, evaluating apoptosis and autophagy readouts, and use in target validation and small-molecule screening workflows; the concentration or dose used in experiments typically depends on the specific experimental design and research objectives.
