Nortadalafil (CAS 171596-36-4) is a demethylated analogue of tadalafil, a selective inhibitor of phosphodiesterase type 5 (PDE5). PDE5 selectively hydrolyzes cyclic GMP (cGMP) within the corpus cavernosum, modulating vasodilation essential for penile erection. By inhibiting PDE5, nortadalafil contributes to sustained cGMP-dependent signal transduction, thus enhancing nitric oxide-mediated vasodilation pathways. Tadalafil displays potent inhibition of PDE5 with an IC50 of approximately 5 nM and marked selectivity over other PDE isoforms (PDE1–4, PDE6). Nortadalafil serves as an analytical reference for pharmacological and biochemical studies on PDE5 inhibitors in erectile dysfunction and pulmonary arterial hypertension research.
