Nicotinamide
Nicotinamide (CAS: 98-92-0) is a small-molecule inhibitor of poly(ADP-ribose) polymerase-1 (PARP-1), an enzyme critical in DNA repair pathways. Under pathological conditions such as ischemia or trauma, PARP-1 hyperactivation depletes cellular NAD+, triggering neuronal cell death. In vitro studies indicate that transporter-mediated uptake of nicotinamide confers protection against PARP-1-induced astrocyte injury. In vivo studies in diabetic rat models demonstrate that nicotinamide administration normalizes hepatic antioxidant enzymes, reducing oxidative stress. Clinical trials have shown dose-dependent upregulation of frataxin expression following sustained nicotinamide treatment, suggesting therapeutic potential in disorders like Friedreich's ataxia.
