Background

EN6 is a small-molecule in vivo autophagy activator that covalently targets cysteine 277 in the subunit of the lysosomal enzyme.277 EN6-mediated modifications couple the enzyme to uncoupling, resulting in inhibition of signaling, increased lysosomal acidification, and activation of autophagy.EN6 also removes aggregates (the causative agent of frontotemporal dementia) in a lysosome-dependent manner.
