Digitonin
Digitonin (CAS No.: 11024-24-1) is a steroidal saponin glycoside derived from Digitalis purpurea widely used in biomedical research as a natural, cholesterol-binding detergent that selectively permeabilizes cellular membranes. By interacting with membrane cholesterol, digitonin disrupts lipid bilayer integrity and increases cell permeability without substantially impairing mitochondrial function, enabling controlled access to intracellular compartments for biochemical and pharmacological studies. This mechanism underlies its utility in facilitating the intracellular delivery and uptake of chemotherapeutic agents, as demonstrated by enhanced accumulation and cytotoxicity of platinum-based compounds such as cisplatin analogs in cultured cancer cells and increased tumor uptake and efficacy of carboplatin in preclinical models without exacerbating systemic toxicity. Digitonin is therefore commonly applied in studies of membrane dynamics, organelle function, and drug transport, as well as in protocols requiring selective permeabilization for enzyme assays or mitochondrial analyses. Its functional activity is typically observed at low micromolar concentrations in vitro, although optimal conditions vary depending on cell type and experimental design, and dosing in animal models is adjusted according to specific study objectives.
