Background

AZD0156 (CAS number: 1821428-35-6) is an orally bioavailable, potent and selective small-molecule inhibitor of ataxia telangiectasia mutated (ATM) kinase, a serine/threonine kinase within the phosphatidylinositol 3-kinase-related kinase (PIKK) family. ATM kinase contributes critically to DNA damage response signaling activated by DNA double-strand breaks, thus facilitating DNA repair, checkpoint control, genomic stability, and cell fate decisions. AZD0156 demonstrates sub-nanomolar inhibitory potency against cellular ATM signaling and exhibits over 1000-fold selectivity relative to other PIKK enzymes. In preclinical models, oral AZD0156 administration potentiates antitumor responses induced by double-strand break-generating agents. Currently, AZD0156 is undergoing early clinical trials assessing safety and preliminary efficacy for advanced cancers.
