UNC2025 HCl
UNC2025 HCl (CAS No.: 2070015-17-5) is a synthetic ATP-competitive small molecule classified as a dual-target receptor tyrosine kinase inhibitor, with the TAM family member MERTK and the hematopoiesis-related kinase FLT3 as its primary targets. It is commonly used in biomedical research such as tumor biology and immunobiology, as well as in mechanistic analysis and target validation in early-stage drug discovery; by occupying the ATP-binding site of kinases, it inhibits autophosphorylation of target proteins and downregulates signal transduction, and can be used to dissect the GAS6–TAM axis and FLT3-driven downstream PI3K–AKT, MAPK–ERK, and STAT cascade systems. In relevant cellular systems, inhibition of cell proliferation, induction of programmed cell death, and regulation of apoptotic cell clearance (efferocytosis) can be observed. In vitro biochemical and cellular assays show activity in the subnanomolar to low-nanomolar range, with an approximately one- to two-order-of-magnitude selectivity advantage for MERTK over AXL and TYRO3, including more than 45-fold selectivity over AXL. Studies have also reported favorable pharmacokinetic characteristics, including oral bioactivity and a certain degree of brain exposure, supporting its application in peripheral and central nervous system-related mechanistic studies. Typical research applications include pathway validation and target-dependency studies in leukemia and various solid tumor cell lines, signal transduction analysis in macrophages or microglia, and pharmacodynamic/pharmacological biomarker evaluation in xenograft or other animal models; it can also be used in primary/secondary screening platforms for lead compound discovery and combination interventions, with the specific concentrations or dosing levels used in experiments typically determined according to the experimental design and research objectives.
