TPE-MI (Tetraphenylethenemaleimide) is intrinsically non-fluorescent until it binds to thiols via maleimide.TPE-MI fluorescence is activated upon labeling of free cysteine thiols, which are normally buried in the core of globular proteins exposed upon unfolding.TPE-MI can be used to measure cellular unfolded protein load.TPE -MI can report imbalances in protein homeostasis in the Huntington's disease induced pluripotent stem cell model, as well as in cells transfected with mutant Huntington exon 1 prior to the formation of visible aggregates.TPE-MI also detects protein damage after dihydroartemisinin studies of malaria parasites.
