Ponatinib Hydrochloride
Ponatinib is a third-generation tyrosine kinase inhibitor designed to inhibit BCR‑ABL, especially the T315I mutant that is resistant to other TKIs. It blocks downstream signaling such as STAT5, PI3K/AKT, and RAS/MAPK by occupying the BCR‑ABL ATP-binding site, inducing apoptosis in leukemia cells. It also inhibits multiple kinases, including VEGFR, FGFR, PDGFR, SRC, KIT, and FLT3, and therefore shows antitumor activity in preclinical models of CML, Ph+ ALL, and some solid tumors. Its multi-target activity is also associated with adverse reactions such as arterial thrombosis and cardiovascular events.
