NVP-2
NVP-2 (CAS No.: 1263373-43-8) is a highly potent and selective ATP-competitive inhibitor of cyclin-dependent kinase 9 (CDK9), a key regulator of transcriptional elongation through its association with cyclin T. By targeting the CDK9/CycT complex with sub-nanomolar potency, NVP-2 effectively suppresses RNA polymerase II–mediated transcription, thereby modulating gene expression programs critical for cell survival and proliferation. In addition to its primary activity, NVP-2 exhibits weaker inhibitory effects against related kinases such as CDK1/CycB, CDK2/CycA, and CDK16/CycY in the micromolar range, indicating a high degree of selectivity for CDK9. Functionally, inhibition of CDK9 by NVP-2 leads to rapid transcriptional repression and induction of apoptosis in sensitive cell types, making it a valuable chemical probe for studying transcriptional regulation, oncogenic signaling, and dependency on transcriptional machinery in cancer and other diseases. This compound is widely utilized in cell-based assays and mechanistic studies to investigate CDK9-driven pathways, and its application in experimental systems typically depends on specific study designs and biological contexts.
