Nestoron (CAS No.: 7759-35-5) is a synthetic progestin and a high-affinity agonist of the progesterone receptor, with an EC50 of approximately 10.3 nM. In vitro studies show that Nestoron exhibits significant binding to the calf thymus glucocorticoid receptor, with an ED50 of approximately 56 nM. Long-term use of Nestoron can increase the generation of neurons and oligodendrocytes. In vivo studies indicate that the potency of subcutaneous administration of Nestoron is more than 100-fold higher than that of the oral route, and it does not exhibit androgenic or cationic activity, nor does it bind to sex hormone-binding globulin. In addition, Nestoron did not show uterotrophic activity in ovariectomized rats. Although it shows significant binding to the glucocorticoid receptor, it did not exhibit glucocorticoid activity in vivo.
