Background

Mycro 3 (CAS No.: 944547-46-0) is a small-molecule inhibitor targeting the oncogenic c-Myc signaling axis, classified as a selective disruptor of Myc–Max protein–protein interactions with demonstrated oral activity. It functions by inhibiting the dimerization of c-Myc with its obligate partner Max and consequently suppresses c-Myc-dependent DNA binding and transcriptional activity, thereby interfering with a central regulatory pathway in cancer cell proliferation and metabolism. The compound exhibits selective activity toward c-Myc-driven systems, showing reduced effects on related transcriptional regulators such as AP-1, and preferentially decreases viability in c-Myc-expressing cells compared to c-Myc-deficient counterparts, with inhibitory effects observed in the low micromolar range in cellular assays. In preclinical models, Mycro 3 has demonstrated the ability to reduce tumor growth in pancreatic cancer xenografts and to enhance therapeutic efficacy when combined with standard anticancer agents in prostate cancer models, supporting its utility as a tool compound for investigating Myc-dependent oncogenesis and combination treatment strategies. It is widely applied in biochemical and cell-based assays to study transcription factor regulation, protein–protein interaction inhibition, and oncogenic signaling, with experimental concentrations and dosing regimens varying according to specific model systems and study objectives.
