Miransertib (ARQ-092) is an oral bioavailable, selective, and potent allosteric Akt inhibitor with an IC50 of 2.7 nM for Akt1, 14 nM for Akt2, and 8.1 nM for Akt3, respectively.
Miransertib has a high enzymatic effect on Akt1, Akt2 and Akt3, and can effectively inhibit the activation of Akt and the phosphorylation of downstream target PRAS40. In a large number of cell lines from various tumor types, Miransertib showed strong anti-proliferative activity in cell lines containing PIK3CA/PIK3R1 mutations compared to those with wild-type (wt) PIK3CA/PIK3R1 or PTEN deletion. Miransertib inhibited p-Akt (S473) and p-Akt (T308) in both AN3CA and A2780 cells.
