Background

Lys05 3HCl (CAS No.: 1391426-24-6) is a synthetic small molecule, also known as Lys01 trihydrochloride, classified as a lysosomotropic lysosomal autophagy inhibitor and commonly used as a chemical probe in cell biology and drug discovery; it is derived from a dimeric analog of the chloroquine chemotype and is presented as a water-soluble trihydrochloride form, with the property of preferentially accumulating in lysosomes within cells and increasing their internal pH, thereby interfering with autophagic flux at the stage after autophagosome-lysosome fusion; mechanistically, it can induce accumulation of p62/SQSTM1 and increase the LC3-II/LC3-I ratio, and an increased burden of autophagy-related vesicles has been observed in tumor tissues under xenograft conditions; compared with hydroxychloroquine, it generally shows more pronounced lysosomal accumulation and autophagy-inhibitory effects in vitro; it displays low micromolar in vitro activity in multiple human cell lines (such as 1205Lu、C8161、LN229、HT-29, etc.) and is commonly characterized through cell viability and autophagy marker readouts; it is mainly used to dissect lysosome-dependent signaling pathways and cellular stress responses, to controllably modulate autophagic flux in cellular and animal models to support target validation, and to perform combinatorial mechanistic or phenotypic screening with molecules acting on autophagy or lysosomal function; the specific concentration or dose used in experiments typically depends on the research objective and experimental design.
