Ilaprazole (CAS No.: 172152-36-2) is a novel proton pump inhibitor (PPI) that irreversibly inhibits the H+/K+-ATPase on gastric parietal cells, significantly reducing gastric acid secretion. It is mainly used to treat acid-related diseases such as dyspepsia, peptic ulcer, gastroesophageal reflux disease, and duodenal ulcer. Studies show that Ilaprazole has significant in vitro antibacterial activity against Helicobacter pylori (H. pylori), with a minimum inhibitory concentration (MIC50/MIC90) of approximately 8 μg/mL, and exhibits synergistic effects when combined with clarithromycin. A quadruple regimen (Ilaprazole, amoxicillin, clarithromycin, and potassium bismuth citrate) shows significant synergistic bactericidal activity. Its mechanism involves disrupting the bacterial membrane potential (ΔΨ), enhancing antibiotic efficacy, and continuous passaging did not induce resistance, providing a theoretical basis for optimizing H. pylori eradication strategies. In in vitro studies, Ilaprazole inhibits H+/K+-ATPase in rabbit parietal cell preparations with an IC50 of approximately 6 μM. In addition, Ilaprazole is also an effective T-cell-originated protein kinase (TOPK) inhibitor, with potential value in antitumor research. In cell and animal models, Ilaprazole is commonly used to evaluate its efficacy in inhibiting gastric acid secretion and anti-ulcer effects, and the doses or concentrations used in experiments usually depend on the experimental design and research objectives.
