Background

Cholesterol in blood is mainly transported as lipoproteins, including HDL-C, LDL-C, and VLDL-C. HDL-C, rich in ApoA-I, facilitates reverse cholesterol transport and exhibits antioxidant and anti-inflammatory properties; reduced levels correlate with increased atherosclerotic cardiovascular disease (ASCVD) risk. LDL-C, containing ApoB-100, holds the highest cholesterol content in plasma and is a primary target for lipid intervention. VLDL-C, synthesized in the liver, transports endogenous triglycerides and converts to LDL-C; its elevation indicates metabolic disorders. Accurate measurement of HDL-C and LDL/VLDL-C is vital for disease and health management.

This kit utilizes chemical precipitation and enzymatic reactions for efficient lipoprotein separation and quantification. Following precipitation, cholesterol esters in HDL-C or LDL/VLDL-C fractions are hydrolyzed by cholesterol esterase and cholesterol oxidase to produce hydrogen peroxide, which reacts with Amplex Red via horseradish peroxidase to generate a fluorescent signal proportional to cholesterol content. Compatible with serum, plasma, and tissue samples, the kit offers high sensitivity and a wide linear range, supporting both fluorescence and absorbance detection for basic research and high-throughput screening.
