Fexinidazole
Fexinidazole (CAS No.: 59729-37-2) is a potent nitroimidazole compound mainly used in biomedical research and drug discovery, and has attracted broad attention due to its antiparasitic properties. This compound exhibits significant anti-trypanosomal activity in various in vitro models, particularly against multiple strains of Trypanosoma brucei, the causative agent of human African trypanosomiasis, with inhibitory concentrations (IC50) in the low micromolar range. Studies have also shown that Fexinidazole has certain activity against other protozoan parasites such as Trypanosoma cruzi, Tritrichomonas foetus, Trichomonas vaginalis, Entamoeba histolytica, and Leishmania donovani. During metabolism, this compound is converted into biologically active sulfone and sulfoxide forms, and its antiparasitic effects have been validated in various in vitro models; for example, it shows half-maximal effective concentrations (EC50) in the low micromolar range in both the promastigote and amastigote stages of Leishmania donovani. In mouse infection models, Fexinidazole can reduce parasite burden and improve host survival, with the doses used typically adjusted according to the specific experimental design and research objectives. As an important tool compound, Fexinidazole is used in drug screening and related mechanistic studies, providing important support for the understanding of parasitic infections and the development of countermeasures.
