Estetrol
Estetrol (CAS No.: 15183-37-6) is a naturally occurring estrogen uniquely synthesized by the human fetal liver during pregnancy and is classified as a selective nuclear estrogen receptor modulator. In the context of biomedical research and drug development, this compound exhibits its unique selective modulatory effects through interactions with the estrogen receptors ERα and ERβ. Although its binding affinity for ERβ is one-fourth that for ERα, this receptor-differential binding forms the basis of its selective modulatory effects. Estetrol can induce the expression and activity of endothelial nitric oxide synthase (eNOS), thereby enhancing nitric oxide (NO) synthesis in endothelial cells. In in vitro studies, its activity levels typically range from nanomolar to micromolar concentrations, depending on the specific experimental conditions and research objectives. Estetrol has estrogenic effects on the endometrium and central nervous system, but shows antagonistic effects in breast tissue, which is a key mechanistic distinction. These properties make Estetrol a valuable candidate compound for exploring contraception and menopausal hormone replacement research. In addition, exploratory studies suggest that it may reduce thrombosis risk at low concentrations, although its precise role in this pathway still requires further investigation. This compound is used in various cell and animal model studies to elucidate its biological effects and drug development potential. Experimental designs and dosing regimens are tailored to specific scientific questions, making it an insightful tool for studying estrogen pathways and their applications in human health and disease models.
