Background

Eganelisib (CAS No.: 1693758-51-8) is a synthetic small molecule derived from medicinal chemistry optimization and a highly selective phosphoinositide 3-kinase γ (PI3Kγ) inhibitor, commonly used as a tool compound in studies of signal transduction, macrophage biology, and the tumor microenvironment; its activity is primarily localized to the γ isoform among class I PI3K isoenzymes, and it exhibits inhibitory and functional effects in the nanomolar to low micromolar range in biochemical and cellular systems, while typically showing more than 100-fold selectivity over other class I isoforms and low off-target interference against a broad range of protein and lipid kinases as well as other receptor families under micromolar-level multi-target screening conditions. Mechanistically, its blockade of PI3Kγ can inhibit activation of the PI3K–AKT pathway, manifested as downregulation of AKT phosphorylation at the Ser473 site in various cellular contexts; functionally, it can limit chemokine-driven migration of bone marrow-derived macrophages and has been used to dissect myeloid cell-mediated microenvironmental regulation. In addition to pathway inhibition, in multidrug resistance-related studies, this compound can be used to investigate P-glycoprotein-mediated drug efflux and substrate accumulation, providing quantifiable modulatory value for the intracellular levels and in vitro effects of co-administered substrates. Its typical research uses include target validation, pathway mapping, pharmacodynamic and target occupancy assessment, myeloid cell phenotypic analysis, and hypothesis testing of combination strategies in early discovery models such as cell culture and mouse xenografts; the concentrations or doses used in specific experiments are usually determined according to the experimental design and research objectives, supporting biomedical research and drug development scenarios such as mechanism elucidation, phenotypic screening, and optimization of combination strategies.
