Background

EC330 (CAS No.: 2016795-77-8) is a steroidal small-molecule pathway probe classified as a leukemia inhibitory factor (LIF) signaling antagonist, suitable for life science research contexts such as tumor biology and cytokine biology; by binding to the LIF receptor (LIFR) and blocking the LIF-dependent signal transduction axis, it inhibits the pro-growth and pro-migration effects of this pathway on tumor cells, and shows selective cytotoxicity and inhibition of migratory phenotypes in multiple tumor cell lines (including the NCI?60 panel), with in vitro activity typically in the low nanomolar to submicromolar range. In addition to pathway antagonism, EC330 can induce ferroptosis-related responses in vitro characterized by decreased intracellular glutathione and reduced glutathione peroxidase activity, providing a mechanistic entry point for analyzing redox-vulnerable states and lipid peroxidation stress. This molecule contains a terminal alkyne group and can be used for bioorthogonal labeling and target occupancy studies via copper-catalyzed azide-alkyne cycloaddition, supporting protein enrichment, binding-site identification, and target validation in chemical biology workflows. Typical applications include target validation and phenotypic screening in cell models with highly activated LIF backgrounds (such as breast-derived cancer cell models), as well as exploratory evaluation of pharmacological hypotheses under LIF overexpression conditions in xenograft models; the specific concentration or dose used in experiments depends on the experimental design and research purpose, and it is suitable for in vitro mechanistic analysis and screening studies.
