EB-3D
EB-3D (CAS No.: 1839150-63-8) is a potent and selective inhibitor of choline kinase alpha 1 (ChoKα1), a key enzyme involved in phosphatidylcholine biosynthesis that is frequently upregulated in aggressive cancer phenotypes. With inhibitory activity in the low micromolar range, EB-3D effectively reduces phosphocholine production and disrupts lipid metabolism, leading to marked suppression of cellular proliferation, migration, and invasion in multiple cancer cell models, including breast cancer and T-cell leukemia systems. Mechanistically, EB-3D has been shown to induce apoptosis, promote endoplasmic reticulum stress, and trigger early activation of AMP-activated protein kinase (AMPK), resulting in downstream inhibition of the mTORC1 signaling pathway and modulation of associated effectors such as 4E-BP1, p70S6K, S6 ribosomal protein, and GSK3, ultimately impairing protein synthesis and cell viability. In addition, EB-3D has been reported to enhance chemosensitivity in leukemia cell lines and to induce cellular senescence. In vivo studies using murine breast cancer models have demonstrated its ability to reduce tumor burden and limit metastatic dissemination at experimentally determined dosing regimens. This compound is widely utilized as a chemical probe for investigating choline metabolism, AMPK–mTOR axis regulation, and lipid-associated signaling pathways in oncology-focused research, with experimental concentrations and treatment conditions varying according to specific study designs.
