DUB-lN-1 (CAS No.: 924296-18-4) is a ubiquitin-specific protease/deubiquitinase inhibitor, mainly used in research on USP8-related deubiquitination processes and ubiquitin-specific protease pathways. The IC50 of this compound for inhibition of USP8 enzymatic activity is 0.85 μM, while it has weak or no activity against USP7 enzymatic activity (IC50 > 100 μM), and can be used for USP8/USP7 selectivity comparison. In HCT116 and PC-3 cell viability assays. The IC50 range of this compound and its analogs is 0.5–1.5 μM; in ESCC cell viability assays, the IC50 is 1.58–2.14 μM, and it is involved in studies of apoptosis, autophagy, and G2/M phase cell cycle arrest in KYSE-450 and KYSE-30 models.
