DSPE-DBCO
DSPE‑DBCO (CAS No.: 2097415-18-2) is a click lipid in which a DBCO (dibenzocyclooctyne) group is directly conjugated to the headgroup of the DSPE phospholipid (1,2‑distearoyl‑sn‑glycero‑3‑phosphoethanolamine); the small molecule itself does not contain a PEG chain. As a classic pharmaceutical excipient, DSPE offers good emulsification and solubilization capacity and is a commonly used scaffold material in sustained/controlled‑release formulations such as liposomes, lipid emulsions, and nanoparticles; on this basis, introducing DBCO at the polar headgroup can endow the carrier with controllable bioorthogonal “clickable” functional sites. DSPE‑DBCO retains DSPE’s ability to insert into lipid bilayers and stabilize liposomes/nanoparticles, while also enabling copper‑free SPAAC reactions in aqueous phase with azide (N₃)‑containing drugs, fluorescent probes, ligands, etc., allowing post‑modification of liposome or nanoparticle surfaces to construct lipid formulations with targeting, environment‑responsive, or tracing functions, making it a useful tool molecule for developing new targeted drug delivery systems and functionalized nanocarriers.
