Crotamiton
Crotamiton (CAS No.: 483-63-6) is a synthetic ectoparasiticide and antipruritic agent widely utilized in dermatological research and drug discovery contexts for its dual activity in alleviating itch and eradicating scabies-causing parasites. Functionally, it has been identified as an inhibitor of transient receptor potential vanilloid 4 (TRPV4) channels, suppressing TRPV4-mediated currents in a calcium-dependent manner and modulating pruritus-associated signaling pathways. In vitro and cellular studies demonstrate that crotamiton inhibits histamine- and chloroquine-induced calcium influx through pathways involving H1 receptor/TRPV1 and MRGPRA3/TRPA1 signaling, as well as attenuating calcium responses in primary dorsal root ganglion neurons, indicating a broader regulatory role in sensory neuron activation. In vivo, crotamiton reduces pruritus-related behaviors in murine models induced by TRPV4 agonists and other pruritogens such as histamine, serotonin, and PAR2 agonists, highlighting its relevance for translational itch research.
