Casin (CAS 425399-05-9) is a selective inhibitor targeting Cdc42, a member of the Rho GTPase family involved in regulating actin and microtubule organization, cellular polarity, and cell adhesion. Casin inhibits the active, GTP-bound state of Cdc42 at an IC50 of approximately 2 μM. In vitro, Casin treatment reduces elevated levels of active Cdc42 observed in aged hematopoietic stem cells (HSCs) toward youthful levels, restoring cellular polarization phenotypes. In animal studies, Casin administration reversed age-associated polarity defects in aged HSCs. Casin is under investigation in preclinical aging and stem cell research.
