Background

Batoprotafib (CAS No.: 1801765-04-7) is a synthetic, orally available allosteric small molecule, classified as a signal transduction research tool that selectively inhibits Src homology 2 domain-containing protein tyrosine phosphatase 2 (SHP2, PTPN11). SHP2 serves as a cytosolic regulator that couples receptor tyrosine kinase (RTK) signaling to the RAS–MAPK cascade and related networks; Batoprotafib shows nanomolar biochemical inhibitory potency against SHP2 and high selectivity over multiple classes of kinases and phosphatases, facilitating precise analysis of SHP2-dependent pathways in complex systems. In cellular models, this molecule can reduce ERK phosphorylation and inhibit proliferation driven by RTKs and under BRAF-mutant backgrounds, typically exhibiting functional activity ranging from low nanomolar to low micromolar levels; in mechanism- and pharmacology-driven early in vivo studies (such as xenograft mouse models), associations can be observed between modulation of pathway output and restriction of tumor growth, thereby supporting its application in studies of oncogenic signaling axes, target validation, and analysis of adaptive feedback related to sensitivity/resistance. Batoprotafib is widely used to explore PTPN11 biology, evaluate feedback and bypass activation within the RTK–RAS–MAPK axis, and assess combination strategies with pathway inhibitors (such as RAF or MEK inhibitors) in vitro and in vivo; in drug discovery workflows, it is also suitable as a standard control or reference compound for biochemical and cellular screening, organoid platforms, and pharmacokinetic–pharmacodynamic correlation studies.
