Acridine Orange 10-Nonyl Bromide
Acridine Orange 10-Nonyl Bromide (NAO), a lipophilic cationic derivative of acridine orange, readily traverses live-cell membranes and binds specifically to cardiolipin, an anionic phospholipid enriched in the inner mitochondrial membrane. NAO exhibits green fluorescence (Ex/Em ~496/524 nm). Notably, localized probe accumulation promotes oligomeric stacking with cardiolipin, inducing a spectral red shift with peak excitation/emission at ~450/640 nm.
Unlike potential-sensitive probes such as Rhodamine 123 and JC-1, NAO accumulation is relatively independent of mitochondrial membrane potential (ΔΨₘ), enabling robust quantification of mitochondrial mass and assessment of inner membrane integrity. Consequently, NAO is widely employed for live-cell mitochondrial morphotyping, monitoring cardiolipin dynamics during apoptosis, flow cytometric analysis of mitochondrial content, and staining Gram-negative bacteria.
