(±)-Blebbistatin (CAS: 674289-55-5) is a cell-permeable small molecule inhibitor selectively targeting non-muscle myosin II (NM II) ATPase. It preferentially binds the myosin-ADP-Pi intermediate complex, delaying phosphate release and consequently preventing actin-activated ATPase activity and myosin II-mediated contraction. Blebbistatin inhibits several isoforms of myosin II across multiple species with IC50 values from approximately 0.5 to 5.0 μM, but exhibits significantly weaker inhibition towards smooth muscle myosin II and myosin isoforms I, V, and X. This inhibitor is widely applied in cellular research to reversibly modulate cytoskeletal dynamics, cell migration, adhesion, and morphology.
