Background

1,8-ANS (CAS No.: 82-76-8) is a widely used environment-sensitive fluorescent probe in biochemical and biophysical research, particularly for interrogating protein structure, conformational dynamics, and hydrophobic surface exposure. This small-molecule dye exhibits low fluorescence in aqueous solution but undergoes a marked increase in intensity and a characteristic blue shift upon binding to hydrophobic regions of proteins, enabling sensitive detection of folding states, ligand-induced conformational changes, and membrane protein interactions. It displays excitation and emission maxima near 350 nm and 520 nm in solution, respectively, with emission shifting to shorter wavelengths upon protein association, and has been applied to characterize binding interactions such as those with fatty acid binding proteins, where affinities are typically observed in the low micromolar range. In addition to its role as a biophysical probe, 1,8-ANS functions as a competitive inhibitor of thyroxine-binding globulin, interfering with triiodothyronine binding and facilitating radioimmunoassay workflows by increasing the availability of free hormone for antibody recognition, with activity reported in the micromolar affinity range. Mechanistically, its utility derives from selective partitioning into low-polarity microenvironments and reversible occupancy of hydrophobic binding pockets, making it valuable for screening ligand–protein interactions and assessing protein stability in vitro. It is broadly employed across protein folding studies, drug discovery assays targeting hydrophobic binding sites, and the evaluation of biomolecular interactions in cell-free systems, with experimental concentrations adjusted according to assay format, protein targets, and detection sensitivity requirements.
